چکیده

The requirement for a group of normal serum factors, collectively designated as complement, for injury or destruction of cells sensitized with antibody has been recognized for more than 70 years (1). The participation of complement in immune destruction of erythrocytes (immune hemolysis) has been most intensively studied, primarily because of technical feasibility. Considerable attention has also been given to the destruction of Gram-negative bacteria by antibody and complement (immune bacteriolysis). Although some evidence is available that normal serum factors resembling complement also participate with antibody in the killing of mammalian cells other than erythrocytes (immune cytotoxicity) (2-13), this activity of complement has been least dearly defined, Investigation of the phenomenon of immune cytotoxicity is of fundamental biological interest from the standpoints of general mechanisms of cellular injury and of specific mechanisms of tissue damage in hypersensitivity states. Although an ultimate aim of such investigation is elucidation of mechanisms of immune injury of human cells in ~ivo, it appeared desirable initially to establish in vitro a model system in which factor requirements and reaction sequences could be studied with greater facility. The model system consisted of freshly isolated normal human amnion cells, rabbit anti-human amnion antibody and normal human serum. The amnion cell was selected because of the desire to establish the model with normal fixed-tissue cells of human origin, the ready availability of sterile human placentas and the relative ease with which suspensions of amnion cells could

STUDIES ON IMMUNE CELLULAR INJURY II. FUNCTIONAL ROLE OF Ctl ESTERASE IN IMMUNE CYTOTOXICITY* BY IRWIN

I t was demonstrated in the preceding paper (1) that incubation of primary isolates of normal human amnion cells with specific rabbit antibody and normal human serum resulted in immune cellular injury as measured by two independent criteria of cytotoxicity. I t was further shown that the factors in normal human serum which were required in addition to specific antibody for production of cytotox...

Studies on Immune Cellular Injury

A model system for the investigation of immune cellular injury in primary isolates of a fixed-tissue cell of human origin has been described, using metabolism-inhibition and uptake of trypan blue as independent criteria of cytoxicity. Cytotoxic effects on human amnion cells were produced by the combination of specific rabbit antibody and factors in normal human serum which were indistinguishabl...

Assessment of protection against SARS-CoV-2 infection by measuring antibody titer, yes or no!

Dear Editor The current epidemic of the novel Coronavirus disease 2019 (COVID-19) risen from Wuhan, China, turned into a worldwide concern because of its incubation period (2-14 days) and its high transmission rate. The first cases of the infection were reported in December 2019 in Wuhan with symptoms like pneumonia for an unknown reason. Very soon it was known as a novel kind of Coronavirus o...

Functional Heterogeneity of Lymphocytec

Infections produced in adult mice by the neurotropic Armstrong strain of lymphocytic choriomeningitis (LCM)' virus can result in either of two contrasting outcomes, each of which depends on the route of virus administration . Inoculated intracerebrally (i .c .), any dose of infectious virus replicates predominantly in neural membranes, with little or no extension to parenchymal cells of the cen...

Acute lung injury in rat caused by immunoglobulin A immune complexes.

Mouse IgG and IgA, with reactivity to dinitrophenol conjugated to carrier protein, have been isolated from myeloma proteins by means of a variety of affinity techniques. The IgA was predominantly in the dimeric form. The in vitro and in vivo biological activities of IgA-containing immune complexes were assessed in the rat. IgA-containing immune complexes were demonstrated, in a dose-dependent m...
